    Abstract:
    Objective: To investigate the mechanism of treating chronic wounds in rats by using Zhuang medicine Choerospondias axillaris bark. Methods: The whole layer skin defection 40 rats were used and randomly divided into 4 groups:normal group, model group, MEBO group, choerospondias group. The normal group and model group were given external deposited normal saline external application. MEBO group used MEBT/MEBO. Choerospondias group was given external deposited choerospondias axillaris bark water decoction external application. Detected cell ultrastructure, PTEN, VEGF mRNA after 12 d. Results: The organelle structure in MEBO group and choerospondias group were restored. In 12 d, PTEN in MEBO group and choerospondias group were lower, VEGF were higher than 1d (P<0.05). PTEN in MEBO group and choerospondias group were lower, VEGF were higher than model group (P<0.05). MEBO group had no difference with choerospondias group (P>0.05). Conclusion: Choerospondias axillaris bark water decoction could remarkably improve the cell ultrastructure in chronic wounds, regulate the PTEN signal conduction effect, and promote wound healing.
    2  Effects of biochanin A on Hela cell proliferation, migration and Bcl-2 and Bax gene expression in cervical cancer

    Abstract:
    Objective: The study is to investigate the regulation of Biochanin A on proliferation and migration of cervical cancer cells and its possible molecular mechanism. Methods: Hela cells were cultured in vitro and treated with different concentrations of Biochanin A. CCK-8 assay was used to detect the effect of Biochanin A on the proliferation of cervical cancer cells, while the Scratch assay was used to detect the effect on cell migration. Besides, the expression of Bcl-2 /Bax mRNA was detected by RT-PCR. Results: Biochanin A could inhibit the proliferation and migration of Hela cells in a dose-dependent and time-dependent manner. At 10,0,40,0,160 μmol/L of Biochanin A, the inhibition rate of Hela cells increased significantly after 24,8 h, significantly higher than that of the control group. The expression of pro-apoptotic protein Bax increased, while the expression of anti-apoptotic protein Bcl-2 decreased in a dose-dependent manner. Conclusion: Hummus A can inhibit the proliferation and migration of Hela cells in cervical cancer, and its mechanism may be related to the down-regulation of Bcl-2 and up-regulation of Bax.

    Abstract:
    Objective: To investigate the effects of ivabradine on Notch and NF-kappa B signaling pathway in myocardial cells of rats with myocardial infarction. Methods: The model of myocardial infarction was established by ligating the left anterior descending coronary artery. The surviving rats were randomly divided into model group (MI group, n=8) and treatment group (IVA group, n=8). Rats with the same location but without ligation of the left anterior descending coronary artery were used as control group (CON group, n=8). IVA was administered for 28 d. Hemodynamic and cardiac function indexes of all rats were measured:heart rate (HR), systolic pressure (SBP), diastolic pressure (DBP), mean arterial pressure (MAP), left ventricular systolic pressure (LVSP), left ventricular end-diastolic pressure (LVEDP) and maximum rate of increase and decrease of left ventricular internal pressure (+dp/dt); left ventricular mass index, left ventricular cross-sectional diameter and infarct area; The expression of Notch signaling pathway components (Notch-1, Dll-4, Hes-1) in rat cardiomyocytes was detected by PT-PCR, and the expression of DICD-1 and P65 protein was detected by western-blot. One-way ANOVA was used for comparison between groups, and SNK was used for comparison between groups. Results: SBP, DBP, MAP, LASP, LVEDP and (+dp/dt) in MI group were lower than those in control group (P<0.05), while IVA was higher than those in MI group (P<0.05). Left ventricular mass index and left ventricular sectional diameter in MI group were significantly higher than those in control group (P<0.05), but lower than those in IVA group (P<0.05). The expression of Notch-1 in MI group was significantly higher than that in control group (P<0.05), but lower than that in IVA group (P<0.05). There was no significant difference in the expression of Dll-4 and Hes-1 between the three groups (P>0.05). The expression levels of NICD-1 and P 65 in MI group were significantly higher than those in CON group (P<0.05), but lower than those in IVA group (P<0.05). Conclusion: IVA may improve cardiac function and inhibit ventricular remodeling in rats with myocardial infarction through Notch and NF-kappa B signaling pathways.

    Abstract:
    Objective: To observe the Levels of serum CORT, ACTH and CRH in SHR rats, the expression of GR, MR and CRH mRNA in hypothalamus and GR, MR mRNA in hippocampus of the ADHD model animal SHR rat, and to explore the HPA axis function and related factors in SHR rats. Methods: Junior SHR rats were used as ADHD model group and WKY rats as control group. The levels of CORT, ACTH and CRH in serum were detected by ELISA, and the mRNA expressions of GR, MR, CRH in hypothalamus and GR, MR in hippocampus were detected by RT-PCR. Results: The levels of serum CORT and ACTH in SHR rats were significantly lower than those in WKY rats (P<0.01), and the levels of serum CRH were significantly higher than those in WKY rats (P<0.01). GR and MR mRNA expression levels in hypothalamus and hippocampus were significantly higher than those in WKY rats (P<0.01), while CRH mRNA expression in hypothalamus was significantly lower than that in WKY rats (P<0.01). Conclusion: The HPA axis of SHR rats showed hyporesponsiveness, which may be related to CRH priming disorder and high expression of GR and MR mRNA in hypothalamus and hippocampus.
    5  Expression of MicroRNA-188 in gastric carcinoma and its effect on the proliferation and apoptosis of gastric cancer cells

    Abstract:
    Objective: To investigate the expression of microRNA-188 in tissues and blood of gastric cancer patients, and to study the effect of microRNA-188 on the proliferation and apoptosis of human gastric cancer cell line MGC-803 and its mechanism. Methods:Gastric cancer tissues and adjacent tissues were collected from 10 patients with gastric cancer. Quantitative real time PCR(qPCR) was used to detect the expression levels of microRNA-545, microRNA-575, microRNA-1269, microRNA-99a, microRNA-452 and microRNA-188. The levels of six microRNAs in blood samples were detected by qPCR in healthy volunteers and patients with gastric cancer. The microRNA-188 mimics, microRNA-188 AMO and their NCs were transfected into human gastric cancer cells MGC-803 using transfection reagent. After 24 h, CCK-8 assay and TUNEL staining were used to detect the cell proliferation and apoptosis. The website online was used to predict the target gene of microRNA-188. qRT-PCR and western blot assays were used to determine whether microRNA-188 had a regulatory effect on SIX1. Results: Compared with adjacent tissues, microRNA-188 was found to markedly decrease in gastric cancer tissues. Compared with healthy volunteers, microRNA-188 was significantly lower in the blood samples of gastric cancer patients. CCK-8 results showed that microRNA-188 mimics significantly inhibited the proliferation of gastric cancer cells, while microRNA-188 AMO could significantly increase the proliferation of gastric cancer cells. TUNEL staining showed that microRNA-188 can promote the apoptosis of gastric cancer cells, and its AMO can significantly inhibit the apoptosis of gastric cancer cells. qRT-PCR and western blot experiments showed that microRNA-188 can significantly reduce the mRNA and protein levels of its target SIX1. Conclusion: The expression level of microRNA-188 is significantly decreased in the tissue and blood of patients with gastric cancer and microRNA-188 plays an important role in regulating the proliferation and apoptosis of gastric cancer cells by inhibiting SIX1.

    Abstract:
    Objective: To observe the bioavailability of subcutaneous injection of nimodipine and oral administration of nimodipine in healthy rabbits and SAH rabbits, and determine whether subcutaneous administration is superior to oral administration and to target serum concentration.Methods:36 adult male NZW rabbits were divided into 6 groups (n=6) by random number table method:oral nimodipine group (5 mg/kg and 15 mg/kg), subcutaneous injection of nimodipine group (2.5 mg/kg, 5mg/kg,15 mg/kg), SAH+ subcutaneous injection of nimodipine group (2.5 mg/kg), plasma concentrations of nimodipine in each group were measured and statistical analysis was performed. Results: Oral administration of 15 mg/kg of nimodipine produced a higher Tmax and increased Cmax, which was also greater than 5 mg/kg. The time to peak plasma concentration (Tmax) observed after subcutaneous administration had an opposite trend, with 15 mg/kg resulting in a lower Tmax and a lower AUC tendency than 5 mg/kg. (2) Compared with oral administration, the average concentration of nimodipine in the subcutaneous group was significantly greater than 7 ng/mL (P<0.01), and the plasma concentration of nimodipine remained above 7 ng/mL for significantly longer than oral administration (P<0.01). (3) The Cmax, Tmax and AUC values of nimodipine in healthy NZW rabbits were not significantly different from those measured by subcutaneous injection of 5 and 15 mg / kg (P>0.05). There was no significant difference between healthy NZW rabbits (12.9±10.0) ng/mL and SAH (11.8±4.6) ng/mL NZW rabbits at the target level of treatment with an average nimodipine concentration of 7 ng/mL measured at 24 h (P>0.05). Conclusion: Subcutaneous administration of nimodipine is superior to oral nimodipine and can maintain the plasma level of nimodipine at 7 ng/mL after 24 h,which can help to study the mechanism of nimodipine in delaying vasospasm after SAH treatment.
    7  The effect of dopamine neurotransmitter expression on sleep disorders in neurological inpatients

    Abstract:
    Objective: To explore the effect of dopamine neurotransmitter expression on sleep disorders in neurological inpatients. Methods: 34 patients with sleep disorders and 30 volunteers from February 2018 to November 2018 in neurology department of our hospital were selected as the study subjects. They were the observation group and the control group. 64 participants stopped taking stimulating drinks such as coffee and alcohol two weeks ago. The sleep quality and depression were measured by Pittsburgh Sleep Quality Index (PSQI) and Hamilton Depression Scale (HAMD). Degree of depression was examined by brain MRI. Two days later, 11C-Raclopril labeled brain dopamine neurotransmitter binding to receptors was scanned by PET/CT. SPSS was used to analyze the correlation between dopamine neurotransmitters and sleep disorders. Results: The HAMD and PSQI scores of the observation group were significantly higher than those of the control group (P<0.05). On PET/CT scans, 11C-Raclopride was mostly located in nucleus dorsal caudate putamen, caudate nucleus and putamen. The dopamine binding ability of nucleus dorsal caudate putamen, caudate nucleus and putamen in the observation group was significantly lower than that in the control group (P< 0.05). Pearson correlation analysis showed that the course of disease in the observation group was negatively correlated with nucleus dorsal caudate putamen, caudate nucleus and putamen (r=-0.754,-0.717,-0.728;P< 0.05), while the scores of HAMD and PSQI were not correlated with nucleus dorsal caudate putamen, caudate nucleus and putamen (P> 0.05). Conclusion: The decrease of dopamine receptor binding in patients with sleep disorders is related to the course of sleep disorders, suggesting that the decrease of dopamine expression and sleep disorders have clinical significance.

    Abstract:
    Objective: To investigate the effect of prednisone and cyclophosphamide combine with ligustrazine injection on immunologic function and other related factors in patients with systemic lupus erythematosus (SLE). Methods: The subjects selected 70 patients with SLE who diagnosed and treated in our hospital from March 2014 to May 2018, divided into control group and observation group randomly, 35 cases in each group. The patients in the control group were treated with prednisone combined with cyclophosphamide, and the patients in the observation group was given intravenous drip of ligustrazine injection on the basis of the control group. Before and after treatment, detected and compared the immunologic indexes (IgG, C3, ANA), matrix metalloproteinases (MMP-3, MMP-9, TIMP1), chemotactic factor (CXCL9, CXCL10, CXCL11) and serum levels of IL-10, PRL, S100 protein and EET between the two groups. Results: Before treatments, the immunologic indexes(IgG, C3, ANA), matrix metalloproteinases(MMP-3, MMP-9, TIMP1),chemotactic factor(CXCL9, CXCL10, CXCL11) and serum levels of IL-10, PRL, S100 protein and EET between the two groups had no statistical significance(P>0.05); After treatments, the immunologic indexes(IgG, C3, ANA), matrix metalloproteinases(MMP-3, MMP-9), chemotactic factor(CXCL9, CXCL10, CXCL11) and serum levels of IL-10, PRL, S100 protein and EET between the two groups had statistical significances(P<0.05). Conclusion:Ligustrazine injection was added to SLE patients on the basis of prednisone combined with cyclophosphamide therapy, it not only could significantly improve the immunologic function of patients, but also improve the levels of matrix metalloproteinases, chemokines and related serum factors, it's worthy of clinical research and application.

    Abstract:
    Objective: To observe the expression levels of the main molecules of peroxiredoxins (Prdxs) protein family (Prdx1, Prdx2, Prdx4 and Prdx6) in women with postmenopausal osteoporosis (PMOP), and to analyze their clinical diagnostic values. Methods: Patients in Hubei Provincial Hospital of Traditional Chinese Medicine and Wuhan Hospital of Traditional Chinese Medicine diagnosed with PMOP from May 2016 to March 2018 were included as the study group (72 cases), postmenopausal women with normal bone mineral density (BMD) in the same period were also collected as the control group (51 cases). Levels of Prdx1, Prdx2, Prdx4 and Prdx6 in plasma were determined by ELISA. mRNA levels of Prdx1, Prdx2, Prdx4 and Prdx6 in peripheral blood mononuclear cells (PBMC) were determined by reverse transcription quantitative polymerase chain reaction (RT-qPCR). The correlations between Prdxs and clinical parameters were analyzed. Receiver operating characteristic (ROC) curves were used to evaluate the diagnostic values of Prdxs for PMOP. Results: Prdx1, Prdx4 and Prdx6 levels in the study group were significantly lower than those in the control group (P<0.05). The mRNA expression levels of Prdx1 and Prdx6 of PBMC in the study group were significantly lower than those in the control group (P<0.05). Several Prdxs protein levels (plasma) or mRNA levels (PBMC) in the study group were significantly correlated with lipid levels or inflammatory markers levels (P<0.05). The area under the curve (AUC) of plasma Prdx6 for diagnosing PMOP was 0.794 (95% CI =0.714-0.874). And the AUC of mRNA relative expression of Prdx6 in PBMC for diagnosing PMOP was 0.725 (95% CI =0.635-0.814). Conclusions:The decreased expression of Prdxs protein family (especially Prdx1 and Prdx6) is closely related to the incidence of PMOP, and the decreased Prdxs protein family may promote the occurrence of osteoporosis through the abnormal lipid metabolism pathway and the increased systemic inflammation pathway. The detections of Prdx6 levels in plasma and PBMC are of good diagnostic values for PMOP.

    Abstract:
    Objective: To observe the clinical efficacy of Shujin external granules fumigation on the tenosynovitis of finger flexor tendons. Methods: 240 cases of finger flexor tendon stenosing tenosynovitis patients according to treatment successively were randomized into warm water group、particle group and traditional Chinese medicine group,80 cases in each one. In the warm water group,we use warm water fumigation with Votalin to treat. In the particle group,the Shujin external particles fumigation with Votalin were applied. Finally,in the Chinese medicine group, herbs fumigation with Votalin were adopted. We examined and recorded the visual analogue scale(VAS)of 3 groups of patients at the begin of the treatment and the end of the 8th week of the treatment. At the same time,the clinical efficacy of 3 groups of patients were recorded at the end of the 8th week of the treatment. Results: At the end of the 8th week of the treatment,the VAS scores were higher in the particle group and the traditional Chinese medicine group than in the warm water group(P<0.05). The clinical efficacy of particle group and the traditional Chinese medicine group were better than that in the warm water group(P<0.05). Besides,there were no significant difference in the VAS scores and the clinical efficacy between the granule group and the traditional Chinese medicine group(P>0.05). Conclusion: Shujin external granules fumigation for the tenosynovitis of finger flexor tendons can achieve the definite clinical efficacy and is worthy of clinical promotion and application.
    11  Relationship between R5 parameters such as pulse shock pulmonary function test and MEFV and severity of chronic cough in children aged 3 to 5 years old

    Abstract:
    Objective: To investigate the relationship between the parameters of pulse oscillating lung function (IOS), airway viscosity resistance (R5), and maximum expiratory flow-volume curve (MEFV), and the severity of chronic cough in children aged 3 to 5 years. Methods: Eighty children with chronic cough who were diagnosed or treated in our hospital from March 2017 to March 2018 were selected as the study group, and 50 healthy children who underwent physical examination in our hospital were selected as the control group. The child's asthma control test (C-ACT) was used to assess the severity of the disease in the child. MEFV test scores in the two groups included the ratio of forced expiratory volume to forced vital capacity in the first second (FEV1/FVC) and peak expiratory flow (PEF). The reactance value (X5) at R5, the resonance frequency (Fres), and the oscillation frequency of 5 Hz was detected by IOS. The relationship between ACT scores and MEFV and IOS indicators was analyzed by Pearson correlation. The receiver operating characteristic (ROC) curve was used to evaluate the diagnostic value of MEFV and IOS indicators for chronic cough. Results: The C-ACT score of the severe group was significantly lower than that of the control group (P<0.05). FEV1/ FVC and PEF were lower in the mild and severe groups than in the control group. The severe group was lower than the mild group (P<0.05). Fres, R5 and X5 in the mild and severe groups were significantly higher than the control group, and the severe group was higher than the mild group (P<0.05). FEV1/ FVC and PEF were positively correlated with C-ACT score (P<0.05). There was a negative correlation between Fres, R5 and X5 and C-ACT score (P<0.05). Fres, R5 and X5 showed significant negative correlations with FEV1/FVC and PEF, respectively (P<0.05). R5 has a self-high ROC value of 0.938, followed by Fres, which is 0.917. And the value of IOS diagnostic indicators are higher than MEFV indicators. Conclusion: In children with chronic cough, FEV1/ FVC and PEF decreased while Fres, R5 and X5 increased, and Fres, R5 and X5 had a higher correlation with the severity of cough symptoms, and ROC analysis also showed that R5 was for children aged 3 to 5 years. Chronic cough has the highest diagnostic value.

    Objective: To investigate the expressions of FZD10 and its effect on gastric cancer cells. Methods: The expression of FZD10 in gastric cancer tissues and adjacent cancer tissues was detected by immunohistochemistry;The gastric cancer cell lines AGS and MGC-803 were transfected with siRNA, and then use Western-blot to analysis the transfection efficiency; The proliferation and invasion ability of gastric cancer cell was analyzed by CCK-8 and wound heal. Results: The expression of FZD10 in gastric cancer was significantly higher than that in adjacent tissues (P<0.05). Compared with the blank control group, the proliferation and cell invasion ability of siRNA transfected with AGS and MGC-803 decreased significantly (P<0.05), while the blank control group and empty control group No significant difference between the negative control groups (P>0.05). Conclusion: The expressions of FZD10 significantly up-regulated in gastric cancer, and silencing its expression will affect the proliferation and invasive ability of gastric cancer cells.
    13  Diagnostic efficacy of combined detection of different tumor markers for primary hepatic carcinoma

    Abstract:
    Objective: To explore the clinical diagnostic value of combined detection of different tumor markers for primary hepatic carcinoma, and to provide the reference for the clinical diagnosis. Methods: 72 patients who were diagnosed with primary hepatic carcinoma were collected as observation group, 65 patients with benign liver disease as benign liver disease group and 80 cases of health examination as healthy control group, the contents of tumor markers alpha fetoprotein(AFP), carcinoembryonic antigen(CEA), carbohydrate antigen-199(CA199), carbohydrate antigen-125(CA125) and carbohydrate antigen-153(CA153) were determined by electrochemiluminescence in all subjects, then the results of five kinds of tumor markers and the positive rates of each index between the two groups were compared, the diagnostic value of separate and combined detection of different tumor markers in primary hepatic carcinoma were analyzed. Results: The values of AFP, CA199 and CA153 in the observation group were higher than the benign liver disease group, the values of AFP, CEA, CA199, CA125 and CA153 in the observation group were higher than the control group, the values of CA199 and CA125 in the benign liver disease group were higher than the control group, the differences were statistically significant (P<0.05). The positive rates of AFP, CEA, CA199 and CA153 in the observation group were higher than the benign liver disease group, the positive rates of AFP, CEA, CA199 and CA125 in the observation group were higher than the control group, the positive rates of AFP, CEA, CA199 and CA125 in the benign liver disease group were higher than the control group, the differences were statistically significant (P<0.05). The sensitivity of combined detection of all indicators for primary hepatic carcinoma was 86.4%, specificity, correct index, misdiagnosis rate and missed diagnosis rate were 86.4%, 89.2%, 75.6%, 13.6% and 10.8% respectively, and the combined detection was higher than the correct index of each index. Conclusion:Combined detection of serum tumor markers AFP, CEA, CA199, CA125 and CA153 can improve the sensitivity and specificity of diagnosis of primary hepatic carcinoma, it has better diagnostic value for primary hepatic carcinoma.
    14  Effect of Gongliuxiao Capsule on sex hormones, tumor markers, inflammatory factors and vascular endothelial function in patients with uterine fibroids

    Abstract:
    Objective: To investigate the effects of Gongliuxiao Capsule on inflammatory factors, tumor markers, sex hormones and vascular endothelial function in patients with uterine fibroids. Methods: 130 patients with uterine fibroids admitted to our hospital from October 2015 to March 2017 were enrolled. According to the treatment, they were divided into single treatment group and combined medication group, with 65 cases in each group.The changes of inflammatory factors, tumor markers, sex hormones and vascular endothelial function were compared before and after treatment in the two groups. Results: There were no significant differences in the levels of sex hormones, inflammatory factors, tumor markers and vascular endothelial function between the two groups (P>0.05). The levels of sex hormones E2, P, FSH and LH, the levels of inflammatory factors TNF-α, MMP-9 and IGF-1, tumor markers CA125, CA19-9 and vascular endothelial function VEGF and b-FGF were significantly lower in the two groups (P<0.05). The levels of E2, P, FSH and LH in the combined drug groups were (154.12±16.20) pmol/L, (9.12±1.02) μg/L, (13.84±1.42) U/L and (12.77±1.36) U/L, respectively,which were significantly lower than the single drug group level (P<0.05). The levels of CA125 and CA19-9 in the combined drug group were (27.89±2.88) U/mL and (28.09±3.04) U/mL, respectively, which were significantly lower than the single drug group (P<0.05). The levels of TNF-α, MMP-9 and IGF-1 in the combined drug group were (24.16±2.63) ng/L, (159.22±16.82)μg/L and (69.75±7.09) μg/L, respectively, which were significantly lower than the single drug group(P<0.05). The levels of VEGF and b-FGF in the combination group were (138.29±14.32) μg/L and (98.59±10.02) μg/L, respectively, which were significantly lower than the single drug group (P<0.05). Conclusion:Mifepristone combined with Gongliuxiao Capsule can alleviate endocrine disorders, relieve inflammatory stress response and improve vascular endothelial function in patients with uterine fibroids. It is worthy of clinical application.

    Abstract:
    Objective: To study the effects of dexmedetomidine combined with dezocine on tumor factors, immune function levels and related serum factors in patients with laparoscopic surgery for ovarian cancer. Methods: To select 120 cases of laparoscopic ovarian cancer treated and planned to undergo laparoscopic radical operation in our hospital were randomly and equally divided into control group and treatment group. The patients in control group were treated with dezocine, the treatment group were treated with dexmedetomidine combined with dezocine. To detect and compare the serum HE4, YKL-40, TSGF, NSE, S100β protein, MDA, SOD and peripheral blood CD3+, CD4+, CD8+ and calculate CD4+/CD8+ before and after operation. Results: Before the surgery, there was no significant difference in serum HE4, YKL-40, TSGF, NSE, S100β protein, MDA and SOD between the two groups(P>0.05). After surgery, there were significant differences between the two groups in serum HE4, YKL-40, TSGF, NSE, S100β protein, MDA and SOD levels and the same group before surgery(P<0.05), the improvement of each index in the treatment group was better than those in the control group, and the differences were significantly(P<0.05). Before the surgery, the differences between the two groups in peripheral blood CD3+, CD4+, CD8+ and CD4+/CD8+ had no significant difference (P>0.05), After surgery, the differences between the two groups and the same group before operation in peripheral blood CD3+, CD4+, CD8+ levels and CD4+/CD8+ were significantly(P<0.05), the improvement of each index in the treatment group was better than that in the control group, the differences were significantly (P<0.05). Conclusion: Dexmedetomidine combined with dezocine can reduce the serum HE4, YKL-40, TSGF, NSE, S100β protein levels in patients with ovarian cancer after laparoscopic operation, reduce the oxidative stress reaction, has smaller effect on cellular immune function.

    Abstract:
    Objective: To study the changes in three-dimensional power Doppler ultrasound features before and after neoadjuvant chemotherapy for cervical cancer and their relationship with malignant molecule expression. Methods: The patients who were diagnosed with cervical cancer and received neoadjuvant chemotherapy in Wuhan Red Cross Hospital between March 2015 and October 2017 were selected as the cervical cancer group, and the patients who received cervical biopsy and were diagnosed with stage I cervical intraepithelial neoplasia by pathological findings during the same period were selected as the control group. Before biopsy, three-dimensional power Doppler ultrasonography was performed to measure VI, FI and VFI; the tissues of cervical cancer group before and after chemotherapy as well as the biopsy tissues of control group were collected to measure the expression of proliferation genes, invasion genes and angiogenesis genes. Results: The VI, FI and VFI levels as well as the Piwil2 gene (Piwil2), CyclinD1, N-Myc downstream regulated gene 3 (NDRG3), CXC chemokine ligand 5 (CXCL5), cathepsin-L (CAT-L), EGF-containing fibulin-like extracellular matrix protein 1 (EFEMP1), angiotensin (Ang)1, Ang2 and angiopoietin-like protein 4 (ANGPTL4) mRNA expression levels in the tissues of cervical cancer group before and after chemotherapy were all significantly higher than those of control group whereas the thrombospondin 2 (THBS2), Smac gene (Smac), large tumor suppressor kinase 1 (LATS1), reversion-inducing-cysteine-rich protein with kazal motifs (RECK) and plasminogen activator inhibitor 1 (PAI-1) mRNA expression levels in the tissues were all significantly lower than those of control group, and the VI, FI and VFI levels as well as the Piwil2, CyclinD1, NDRG3, CXCL5, CAT-L, EFEMP1, Ang1, Ang2 and ANGPTL4 mRNA expression levels in the tissues of cervical cancer group after chemotherapy were all significantly lower than those before chemotherapy whereas the THBS2, Smac, LATS1, RECK and PAI-1 mRNA expression levels were all significantly higher than those before chemotherapy; the VI, FI and VFI levels in cervical cancer tissues were positively correlated with the Piwil2, CyclinD1, NDRG3, CXCL5, CAT-L, EFEMP1, Ang1, Ang2 and ANGPTL4 mRNA expression levels, and negatively correlated with the THBS2, Smac, LATS1, RECK and PAI-1 mRNA expression levels. Conclusion: The changes in the three-dimensional power Doppler ultrasound parameters before and after neoadjuvant chemotherapy for cervical cancer can reflect the changes in proliferation, invasion and angiogenesis gene expression in the lesions.
    17  The correlation of serum ROBO4 and sB7-H4 contents with the pathological features of tumor in patients with ovarian cancer

    Abstract:
    Objective: To study the correlation of serum ROBO4 and soluble B7-H4 (sB7-H4) contents with the pathological features of tumor in patients with ovarian cancer. Methods:The patients with ovarian cancer and the patients with ovarian benign tumor who underwent surgical resection in our hospital between March 2015 and August 2017 were selected, and the contents of ROBO4 and sB7-H4 in serum as well as the expression of ROBO4, B7-H4 and oncogenes in the lesion tissues were determined. Results: ROBO4 content in serum of patients with ovarian cancer was significantly lower than that of patients with ovarian benign tumor whereas sB7-H4 content was significantly higher than that of patients with ovarian benign tumor, ROBO4 mRNA expression in ovarian cancer tissues was significantly lower than that in ovarian benign tumor tissues whereas B7-H4 mRNA expression was significantly higher than that in ovarian benign tumor tissues, and ROBO4 and sB7-H4 contents in serum of patients with ovarian cancer were positively correlated with ROBO4 and B7-H4 mRNA expression in ovarian cancer tissues; the higher the FIGO stage and pathological grade of the tumor, the lower the ROBO4 content and the higher the sB7-H4 content in serum of patients with ovarian cancer; β-catenin, CyclinD1 and MMP9 mRNA expression in ovarian cancer tissues were significantly higher than those in ovarian benign tumor tissues, negatively correlated with ROBO4 and positively correlated with sB7-H4 whereas PTEN and E-cadherin mRNA expression were significantly lower than those in ovarian benign tumor tissues, positively correlated with ROBO4 and negatively correlated with sB7-H4. Conclusion: The changes of ROBO4 and sB7-H4 contents in serum of patients with ovarian cancer are closely related to the changes of clinicopathologic stage and the abnormal expression of oncogenes.
